The very first sound/note of Misirlou is surely the most famous example but it's used all over surf, often to signal the imminent appearance of another guitar line. I've tried to do what it sounds like is going on but it just doesn't sound the same. Any suggestions? Thank you!

It isn't audible in the version you listed, but you an see him doing it. But it is sort of a glissando, I wouldn't quantify it as a glissando as it really isn't musical, it is more of special effect. It is just a really quick slide down the low-E string. It shouldn't be too hard to replicate, but it sure was pretty brilliant when he first did it. It could very much so help Dick determine the tempo and be much easier to start the song.

Yes, less audible in the clip I posted, but I posted that one so that the OP could see it being done.
As for musicality, most glissandos in surf tend to be special effects instead of musical.
